A peptide certificate of analysis, commonly called a COA, summarizes the laboratory information reported for a tested sample. Reading one involves more than finding a single purity percentage. A useful review connects the product name, lot number, analytical method, reported result and testing laboratory to the exact material being evaluated.

What is a certificate of analysis?
A COA is a laboratory document that reports information about a particular sample or batch. Depending on the testing ordered, it may include sample identification, analytical methods, chromatograms, mass data, purity calculations, testing dates and laboratory authorization.
The report should be treated as a record of the sample and methods described on that document. It should not be interpreted as a medical recommendation or as proof of an outcome outside the reported analytical work.
Start with sample and lot identification
First, compare the compound name and lot number on the report with the lot information provided for the product. Lot matching matters because a report for one batch should not automatically be assumed to represent every batch produced later.

Identity and purity answer different questions
Identity testing asks whether the analytical characteristics are consistent with the expected compound. Purity testing estimates how much of the detected material is represented by the principal component under the stated test conditions. A purity percentage by itself does not replace identity information.
The U.S. Food and Drug Administration’s analytical-method guidance distinguishes analytical work supporting identity, quality and purity. Although research-product testing is not the same as a regulated drug submission, the distinction between these measurements remains useful when reading a report.
Understanding HPLC results
High-performance liquid chromatography separates components as they move through a chromatographic system. A report may show a chromatogram with one principal peak and smaller secondary peaks. The reported area percentage is calculated from the detected peaks under the method described by the laboratory.
When reviewing an HPLC report, look for the method name, chromatogram, principal peak, integration table and reported purity calculation. Avoid treating the largest number on the page as meaningful without confirming what it measures.
Understanding mass spectrometry
Mass spectrometry evaluates ions according to their mass-to-charge behaviour and can provide evidence related to molecular identity. NIST describes mass spectrometry as an important analytical-chemistry technique used for compound identification and characterization.
A mass result and an HPLC purity result are complementary rather than interchangeable. One may provide evidence related to identity, while the other describes chromatographic composition under the reported conditions.
Check the laboratory and report date
A useful COA should make the testing source and timing understandable. Look for the laboratory name, report or testing date, sample identifier and any signature or authorization details provided by the laboratory. If these details are unclear, the report may be difficult to connect confidently to a particular sample.
Common COA warning signs
- No lot or sample identifier
- A report that cannot be connected to the displayed product lot
- A purity number without a stated analytical method
- Missing laboratory or testing-date information
- Edited, cropped or unreadable result sections
- Identity and purity being presented as if they were the same measurement
